Shanoa's Story - Connect to Castlevania: OoE for DS.
Sypha's Story - Complete Simon's Story.

Unlock Characters
Aeon - Connect to Castlevania Order of Ecclesia.
Shanoa - Connect to Castlevania Order of Ecclesia.

Unlock "True" Story mode
Complete the story mode with Dracula. This mode is noted by a different Character Select background, a second Aeon fight, and a final boss fight with the Time Reaper.

Unlock Difficulty Levels
Brutal - Complete Story on Hard.
Hard - Complete Story on Normal.
